研究方向
本實驗室研究領域是以腸胃道生理學為主,目前研究計畫主要探討腸道菌透過腸-肝軸如何影響B型肝炎病毒持續性或清除感染之預後,主要目的是釐清其於免疫系統中innate myeloid cell和lymphoid cell所扮演的角色,以及腸道屏障功能的完整性,以期望了解腸道菌及其代謝產物如何影響肝臟免疫反應的完整訊息及機制,藉由此研究將有助於開發預防和治療先天免疫細胞相關肝病新穎和創新的策略,以期望未來可藉由細胞免疫療法來重新改編宿主的抗病毒免疫力,進而達成全面清除B型肝炎病毒,對未來B型肝炎的預防與治療提供新契機及新發現。
本實驗室透過疾病的動物模式及細胞分子層次,利用in vivo及in vitro的實驗來完整及有系統的探討腸-肝軸 (gut-liver axis) 相關研究為主,目前實驗室已建立HBV-HDI mice, Germ free mice, CRISPR-Cas9 TLR4 mice, TLR4 mutant mice, intestinal obstruction等小鼠模式,並配合多種分子生物學的實驗技術,以進一步探討細胞分子的作用機轉。實驗技術包括:
(1) Intestinal, liver function and phenotype assay: ALT, AST, HBsAg, HBeAg, Anti-HBs, Anti-Hbe, Anti-Hbc, HBV DNA assay, bacterial translocation, H&E staining, isolation of total liver parenchymal and nonparenchymal cells and flow cytometry analysis.
(2) Biochemical technology: immunofluorescent staining, surface and intracellular staining for flow cytometric analysis and sorting, fluorescent in situ hybridization, ELISPOT, Adoptive transfer, hydrodynamic injection.
(3) Molecular biotechnology: RNA and DNA extraction, RT-PCR, siRNA knockdown, site directed mutagenesis, plasmid transfection, bacterial 16S rDNA sequencing, Temporal temperature gradient gel electrophoresis (TTGE), Southern Bolt, western blotting, Northern blotting. Gene expression profiles using NanoString or RNAseq.
